"Mamono Hunter Youko ²" is an OVA released by Toho in 1995, produced as the sixth installment of the series in response to the aspirations of fans. Youko has inherited the lineage of the Shadow, which is passed down from father to son, so a fight with Youko is inevitable. The story depicts Youko as she struggles to the end with her friends, torn between love and fighting.

The main character, Mano Youko, is 16 years old and was born on June 1st. She is 158cm tall, 45kg in weight, B: 83cm, W: 56cm, H: 85cm, and blood type A. She is a high school student at Kouka Academy, but also fights as the 108th monster hunter. Her best friend, Ogawa Chikako, is 16 years old and was born on August 8th. She is 160cm tall, 53kg in weight, B: 88cm, W: 62cm, H: 88cm, and blood type A. She is a self-proclaimed monster hunter manager, and tries to lose weight, but is not successful because she is greedy for food. Kanzaki Azusa is 13 years old and was born on March 10th. She is 149cm tall, 38kg in weight, and blood type O. She is an apprentice monster hunter and is good at all kinds of housework. Mano Madoka is Youko's grandmother, born on November 11th, and is the 107th monster hunter. Mano Sayoko is Youko's mother and was originally meant to be the 108th monster hunter, but lost her status when she gave birth to Youko at the age of 16. Mano Haruka is the first monster hunter, born on February 24th, and the ancestor of the Mano family.

Other characters that appear include Shouma, a transfer student from the Demon World, Shelmina, the princess of the Aquatic World, Setuna, a nurse in the Wind World, Gart, a doctor in the Wind World, Colt, the prince of the Wind World, Glenn, the prince of the Fire World, Aimi, who works at a bar in the Fire World, Feriam, the creator of the Dream World, Ibex, the god of light who watches over the monster hunters, Dojaki, a monster from the Earth World, Suikoki, a monster from the Aquatic World, Kouchouki, a monster from the Wind World, King Seed, the king of the Fire World, Will, the prince of the Fire World, Bulhoff, who kidnapped Princess Shelmina, Zeluda, the old woman who showed the way to the youkai children, Omu, the big bird of the Wind World, and Valbem, the demon lord who is trying to rule the Dream World.
